Introduction: Micro RNAs (miRNA) are non-coding RNAs that are approximately 19 – 25 base long. miRNAs are responsible of DNA and protein biosynthesis and function as well as cell regulation and pathogenesis via posttranscriptional regulations of the genes. miRNAs can be used as a tool such as humoral rejection and viral infections. Anti HLA antibodies are associated with humoral rejection. The aim of the study is investigation of correlation between miR142-5p and miR192-5p expression and Anti-HLA antibodies positivity.
Materials and Methods: In this study, we evaluated 50 kidney transplant patients who diagnosed acute humoral rejection. We performed the assay on serum/ plasma samples with using Real-Time PCR methods for detection and quantification. Serum PRA sceen accepted positive when up above 25%. For antibody identification LUMÄ°NEX (solid phase assay) assay was used. (project No: KA16/199)
Results: When we compared with each other’s significant differences in miR142-5p and miR192-5p expressions. Spearman correlation analysis showed that miR142-5p was significantly negatively correlated with miR192-5p (r: -0,33, P: 0,018). When we evaluated PRA positivity and miR142-5p and miR192-5p expression we seen that especially upregulation of miR142-5p associated with anti- HLA class-II antibodies with MFI value >1000. Data were analyzed by use SPSS software 17.0
Conclusion: Our results demonstrated that upregulation of miR142-5p and down regulation of miR192-5p are negatively correlated. Upregulation of miR142-5p and anti-HLA-class-II antibodies are positively correlated. These results suggest that upregulation of miR142-5p and down regulation of miR192-5p are associated with antibody mediated rejection in renal transplant patients.
